    Autism Spectrum Disorder and the Criminal Justice System: An Interdisciplinary Study

    The purpose of this study was to advocate and raise awareness of individuals with autism spectrum disorder (ASD) to SIU law students and to inform communication disorders and science (CDS) students of some basic facts about the criminal justice system in the United States. To accomplish this, an interdisciplinary session was conducted in which three CDS students taught ten law students about characteristics of ASD and three law teacher assistants taught thirteen CDS students about disability law and other intricacies of the justice system. Fifteen question pre-surveys were distributed to the participants upon arrival. CDS students answered questions about court proceedings and defendant rights and law students answered questions about various ASD characteristics. Each group of students were divided into three groups (six groups total) and were taught information pertaining to the questions in the surveys in a collaborative manner. Afterwards, all participants took a post-survey identical to the pre-survey. Post-survey results of the law students increased in accuracy by 19.35% (average 4.7 question increase). Post-survey results of the CDS students increased in accuracy by 15.9% (average 2.38 question increase). The interdisciplinary session succeeded in raising law students’ awareness of autism spectrum disorder and in increasing CDS students’ knowledge of the criminal justice system

    Estimating frequencies of emotions and actions: a web-based diary study

    Mental health questionnaires often ask respondents to report how frequently they experience different emotions. We report two experiments designed to assess the accuracy of these reports and the strategies used to generate them. Each day for 2 weeks, participants in Experiment 1 filled out a web-based emotions-and-activities checklist. Then, they estimated the diary-period frequency of these emotions and activities and indicated how they generated each estimate. In Experiment 2, participants provided frequency estimates and strategy reports, but did not fill out the checklist. We found that (a) the frequency estimates were quite accurate for emotions and activities, (b) participants relied on memory-based strategies (enumeration and direct retrieval) when estimating activity frequencies, but (c) used self-knowledge strategies (personality beliefs and schematic inferences) somewhat more than memory strategies for emotions and (d) the relationship between strategy use and question type was unaffected by diary keeping.     Addressing disparities in pharmacogenomics through rural and underserved workforce education

    Introduction: While pharmacogenomic (PGx) testing is routine in urban healthcare institutions or academic health centers with access to existing expertise, uptake in medically-underserved areas is lagging. The primary objective of this workforce education program is to extend access to didactic, case-based and clinical PGx training for pharmacists serving rural Minnesota and populations experiencing health disparities in Minnesota.Methods: A PGx workforce training program funded through the Minnesota Department of Health was offered through the University of Minnesota College of Pharmacy (COP) to pharmacists working in rural and/or underserved areas in the state of Minnesota. Learning activities included a 16-week, asynchronous PGx didactic course covering PGx topics, a 15-min recorded presentation, an in-person PGx case-based workshop, and a live international PGx Conference hosted by the University of Minnesota COP and attendance at our PGx Extension of Community Health Outcomes (ECHO).Results: Twenty-nine pharmacists applied for the initial year of the program, with 12 (41%) being accepted. Four (33%) practiced in a hospital setting, four (33%) in retail pharmacy, two (17%) in managed care, and two (17%) in other areas. The majority had not implemented a PGx program as part of their practice, although nearly all responded definitely or probably yes when asked if they expected their organization to increase its use of PGx testing services over the next three years. All participants either strongly or somewhat agreed that this program helped them identify how and where to access clinical PGx guidelines and literature and improved their ability to read and interpret PGx test results. Eight participants (67%) strongly or somewhat agreed that they expected to increase the number of PGx consultations in their practice, while ten (83%) strongly or somewhat agreed they would be able to apply what they learned in this program to their practice in the next six months to a year.Discussion: This novel PGx training program focused exclusively on pharmacists in rural and/or underserved areas with a delivery method that could be accomplished conveniently and remotely. Although most participants’ organizations had yet to implement PGx testing routinely, most anticipated this to change in the next few years

    Pathways to Increasing Adolescent Physical Activity and Wellbeing: A Mediation Analysis of Intervention Components Designed Using a Participatory Approach.

    We assessed which intervention components were associated with change in moderate-to-vigorous physical activity (MVPA) and wellbeing through proposed psychosocial mediators. Eight schools (n = 1319; 13-14 years) ran GoActive, where older mentors and in-class-peer-leaders encouraged classes to conduct two new activities/week; students gained points and rewards for activity. We assessed exposures: participant-perceived engagement with components (post-intervention): older mentorship, peer leadership, class sessions, competition, rewards, points entered online; potential mediators (change from baseline): social support, self-efficacy, group cohesion, friendship quality, self-esteem; and outcomes (change from baseline): accelerometer-assessed MVPA (min/day), wellbeing (Warwick-Edinburgh). Mediation was assessed using linear regression models stratified by gender (adjusted for age, ethnicity, language, school, BMI z-score, baseline values), assessing associations between (1) exposures and mediators, (2) exposures and outcomes (without mediators) and (3) exposure and mediator with outcome using bootstrap resampling. No evidence was found to support the use of these components to increase physical activity. Among boys, higher perceived teacher and mentor support were associated with improved wellbeing via various mediators. Among girls, higher perceived mentor support and perception of competition and rewards were positively associated with wellbeing via self-efficacy, self-esteem and social support. If implemented well, mentorship could increase wellbeing among adolescents. Teacher support and class-based activity sessions may be important for boys' wellbeing, whereas rewards and competition warrant consideration among girls

    Individual bacteria in structured environments rely on phenotypic resistance to phage

Available on open access from Public Library of Science via the DOI in this recordData Availability: All relevant data are within the paper and its Supporting Information files.Bacteriophages represent an avenue to overcome the current antibiotic resistance crisis, but evolution of genetic resistance to phages remains a concern. In vitro, bacteria evolve genetic resistance, preventing phage adsorption or degrading phage DNA. In natural environments, evolved resistance is lower possibly because the spatial heterogeneity within biofilms, microcolonies, or wall populations favours phenotypic survival to lytic phages. However, it is also possible that the persistence of genetically sensitive bacteria is due to less efficient phage amplification in natural environments, the existence of refuges where bacteria can hide, and a reduced spread of resistant genotypes. Here, we monitor the interactions between individual planktonic bacteria in isolation in ephemeral refuges and bacteriophage by tracking the survival of individual cells. We find that in these transient spatial refuges, phenotypic resistance due to reduced expression of the phage receptor is a key determinant of bacterial survival. This survival strategy is in contrast with the emergence of genetic resistance in the absence of ephemeral refuges in well-mixed environments.     Nuclear Structure of \u3csup\u3e76\u3c/sup\u3eGe from Inelastic Neutron Scattering Measurements and Shell Model Calculations

    The low-lying, low-spin levels of 76Ge were studied with the (n,n′γ) reaction. Gamma-ray excitation function measurements were performed at incident neutron energies from 1.6 to 3.7 MeV, and γ-ray angular distributions were measured at neutron energies of 3.0 and 3.5 MeV. From these measurements, level spins, level lifetimes, γ-ray intensities, and multipole mixing ratios were determined. No evidence for a number of previously placed levels was found. Below 3.3 MeV, many new levels were identified, and the level scheme was re-evaluated. The B(E2) values support low-lying band structure. The 2+ mixed-symmetry state has been identified for the first time. A comparison of the level characteristics with large-scale shell model calculations yielded excellent agreement

    Glioblastoma may evade immune surveillance through primary cilia-dependent signaling in an IL-6 dependent manner

    Glioblastoma is the most common, malignant primary brain tumor in adults and remains universally fatal. While immunotherapy has vastly improved the treatment of several solid cancers, efficacy in glioblastoma is limited. These challenges are due in part to the propensity of glioblastoma to recruit tumor-suppressive immune cells, which act in conjunction with tumor cells to create a pro-tumor immune microenvironment through secretion of several soluble factors. Glioblastoma-derived EVs induce myeloid-derived suppressor cells (MDSCs) and non-classical monocytes (NCMs) from myeloid precursors leading to systemic and local immunosuppression. This process is mediated by IL-6 which contributes to the recruitment of tumor-associated macrophages of the M2 immunosuppressive subtype, which in turn, upregulates anti-inflammatory cytokines including IL-10 and TGF-β. Primary cilia are highly conserved organelles involved in signal transduction and play critical roles in glioblastoma proliferation, invasion, angiogenesis, and chemoradiation resistance. In this perspectives article, we provide preliminary evidence that primary cilia regulate intracellular release of IL-6. This ties primary cilia mechanistically to tumor-mediated immunosuppression in glioblastomas and potentially, in additional neoplasms which have a shared mechanism for cancer-mediated immunosuppression. We propose potentially testable hypotheses of the cellular mechanisms behind this finding

    Content Validity of the Geriatric Depression Scale in Inpatient Health Care Settings

    The content validity of the Geriatric Depression Scale (GDS) for use in inpatient health care settings was evaluated. Clinical experience has shown that one or more questions may not be appropriate in such settings. These questions ask about behaviors or feelings with which the examinee may not be able to identify with because they have been in an inpatient setting during the past week. Twenty-five Subject Matter Experts (SMEs) evaluated the GDS-30 as to whether each item appropriately assesses the construct of depression for inpatients in a medical care setting. SMEs were identified by an online search of the Florida Division of Medical Quality Assurance database; inclusion criteria are presented. Nineteen SMEs held a Ph.D. and six held a Psy.D. Years of post-licensure practice experience ranged from 10 to 48 years with a mean of 23.12 years (SD = 10.07). Using the Content Validity Ratio, four questions emerged as inappropriate (statistically significant at, or below, an alpha level of .025). The lack of content validity of these questions necessitates their omission when psychometrically assessing depression in elderly inpatients. Recommended revised cutoff values are presented. Utilizing the recommended modification to the GDS presented here should reduce false positives when psychometrically assessing depression in elderly inpatients

    Distinct White Matter Changes Associated with Cerebrospinal Fluid Amyloid-β\u3csub\u3e1-42\u3c/sub\u3e and Hypertension

    BACKGROUND: Alzheimer\u27s disease (AD) pathology and hypertension (HTN) are risk factors for development of white matter (WM) alterations and might be independently associated with these alterations in older adults. OBJECTIVE: To evaluate the independent and synergistic effects of HTN and AD pathology on WM alterations. METHODS: Clinical measures of cerebrovascular disease risk were collected from 62 participants in University of Kentucky Alzheimer\u27s Disease Center studies who also had cerebrospinal fluid (CSF) sampling and MRI brain scans. CSF Aβ1-42 levels were measured as a marker of AD, and fluid-attenuated inversion recovery imaging and diffusion tensor imaging were obtained to assess WM macro- and microstructural properties. Linear regression analyses were used to assess the relationships among WM alterations, cerebrovascular disease risk, and AD pathology. Voxelwise analyses were performed to examine spatial patterns of WM alteration associated with each pathology. RESULTS: HTN and CSF Aβ1-42 levels were each associated with white matter hyperintensities (WMH). Also, CSF Aβ1-42 levels were associated with alterations in normal appearing white matter fractional anisotropy (NAWM-FA), whereas HTN was marginally associated with alterations in NAWM-FA. Linear regression analyses demonstrated significant main effects of HTN and CSF Aβ1-42 on WMH volume, but no significant HTN×CSF Aβ1-42 interaction. Furthermore, voxelwise analyses showed unique patterns of WM alteration associated with hypertension and CSF Aβ1-42. CONCLUSION: Associations of HTN and lower CSF Aβ1-42 with WM alteration were statistically and spatially distinct, suggesting independent rather than synergistic effects. Considering such spatial distributions may improve diagnostic accuracy to address each underlying pathology
